ABSTRACT

Metagenomics is an approach to discover novel lipases. Several genes encoding metagenomic lipases have been identified in metagenomic libraries prepared from various environmental samples. Soil samples used in the present investigation have exhibited diverse physicochemical properties. The physical and chemical properties of soil used in this study were diverse. The soil is classified as granular and sandy clay on the basis of particle size. The chapter discusses the methods of purification of crude deoxyribonucleic acid (DNA) using Sephadex Column and silica membrane based commercial spin column. Metagenomic DNA amplification is performed with GeneAmp polymerase chain reaction (PCR) system. PCR products are purified from primers, polymerases, unincorporated nucleotides, buffer components and salts using the QIAquick PCR purification kit. The chapter discusses the metagenomic library construction and blue-white screening, and analysis of transformation efficiency using non-recombinant plasmid.
