ABSTRACT

Chlorophylls, malondialdehyde (MDA) and total thiols (-SH) were analysed as stress indicators.

Chlorophylls were extracted by homogenising 0.25 g of fresh material with acetone (80%) according to Wellburn (1994). The absorbance was determined at 645 and 663 nm. MDA was analysed by adding 1 mL of reagent containing trichloroacetic acid (15%)-thiobarbituric acid (0.37%)-HCl (0.25 M) and butylated hydroxytoluene (0.01%) to 0.1 g fresh weight of plant material (Heath and Packer, 1968). The absorbance was determined at 535 and 600 nm. Total thiols were determined according to Jocelyn (1987). DTBN (5,5′-dithiobis-(2-nitrobenzoic acid)) in 0.5M phosphate buffer was used for the colorimetric reaction. Absorbance was determined at 412 nm.
