ABSTRACT
Thiophilic adsorption chromatography was introduced by Porath and coworkers in 1985 (Porath et al., 1985). The thiophilic adsorbent (T-gel) displays an immobilized ligand: an aliphatic chain having adjacent, (–SO3-CH2-CH2-S-) sulphone and thioether constituents. Since adsorption of proteins on a T-gel is promoted by antichaotropic salts, it can be readily reversed by depleting those salts in the mobile phase. Such cosolvents have a strong influence on the protein adsorption selectivity (Oscarsson et al., 1997, 1998).
