ABSTRACT

Over the past decade synthetic methodologies and reagents in fluorine chemistry have been developed, especially stereocontrolled synthetic methods, enzymatic resolution to synthesize enantiomers, fluoromethylated reagents, and fluorination reagents. This chapter explains how to prepare difluorinated materials in organic fluorine chemistry. It discusses synthetic method of nucleophilic fluorination of 6,6- difluoroundecane, 6,6-difluoro-2-methyloctadec-7-yne, and (E)-1,1-difluoronon-2-ene and electrophilic fluorination of 2’, 3’, 5’-Tri-O-acetyl-5,5-difluoro-5,6-dihydro-6-m ethoxycytidine. The chapter includes miscellaneous of 2-O-benzyl-2-fluoro-3,4-O-isopropylidene-D-ribo-pentopyranosyl fluoride, difluoromethylenation of 2,5-anhydro-6-O-(tert-butyldimethylsilyl)-1-deoxy-1,1-difluoro-3,4-O isopropylidene-D-ribo-hex-1-enitol, 1,1- difluoro-3-[(trimethylsilyl)oxy]-3-phenylbut-1-ene, and octyl 3,3-difluoroacrylate. It provides introduction to difluoromethyl and related groups such as diethyl 2-(bromodifluoromethyl)-2-methylmalonate, diethyl 2-(difluoromethyl)-2-(methylsulfenyl)malonate, 1- benzyloxy-4,4-difluorobut-2-yne, diethyl (bromodifhioromethyl)phosphonate, 1,3- dibromo-1,1-difluorohexane, 1- bromo-1,l-difluorohexane, etc.
