Figure 10.3 Zeta potential distribution curve obtained from the Malvern Zetasizer Nano-ZS. It suggests that the particles are slightly positively charged.We characterized our formulation for toxicity in vitro prior to in vivo evaluation (Fig. 10.4). MTS assay was performed on RAW 246.7 cells at 24, 48, and 72 h time points. Results from the MTS assay confirmed that our particles are non-toxic. Particles were loaded with a fluorescent FITC-albumin to study their uptake in the intestinal cells. Animals in the control group received FITC-albumin solution and negligible amount of FITC-albumin was found in Peyer’s patches as FITC undergoes significant photo-bleaching in acidic pH of the stomach. Hence, only intact FITC-albumin loaded particles could deliver FITC across Peyer’s patches.
