PCR: Polymerase Chain Reaction

PCR allows in vitro amplification of DNA sequences by repetition of elongation reactions in the presence of specific nucleotides and a DNA polymerase. The technique developed from the discovery of a thermophilous eubacterium living in the hot springs (70 to 75°C) of Yellowstone National Park, Thermus aquaticus, and the subsequent use of its polymerase, which is stable up to temperatures close to 100°C.
