Size distribution is also an important factor that should be taken into account. For traditional fabrication methods, size distribution of particles is very broad, which will bring some limitations for their application: (i) the reproducibility of particles is poor among batches, resulting in poor repeatability of the release behavior. In this case, it is difficult to investigate the relationship between doses and treatment effects; (ii) because the accumulated location of the particles highly depends on the size of the particles, the biodistribution of drug is hardly controlled and side-effects are increased.
