ABSTRACT

C ell-free protein expression systems are becoming more widely used as they allow fast synthesis o f recombinant proteins and an easy manipulation o f reaction conditions. Here, we report on recent advances o f a eukaryotic system based on extracts derived from Spodopterafrugiperda insect cells. We demonstrate the use o f PCRproducts as D N A templates for protein production and how this application speeds up in vitro protein expression and potentially cell-based production processes. We also show the expression o f a variety o f functionally active eukaryotic proteins, including kinases and membrane proteins. Finally, we compare the efficiency o f this improved insect-based in vitro protein expression system to traditionally used eukaryotic cell-free extracts, such as rabbit reticulocyte lysate.
