ABSTRACT

From the first procedure standardized in the early 1960s to its most recent modifications, cell-free expression usingEscherichia coli lysates has evolved into a robust method for protein expression capable o f producing protein in milligram quantities. Free from the constraints o f a cell, the components o f the E . coll·based cell-free system can be modified easily to custom­ ize its use for studying all aspects o f protein expression. In this chapter we will discuss the latest advances in E . ca/i-based cell-free protein synthesis focusing on the batch method and its use in high-throughput and structural applications.
