ABSTRACT

C ancer remains one o f the leading causes o f death worldwide. The identification o f new useful and specific biomarkers is necessary in order to detea it earlier. We used cancer immunomics to search and identify proteins o f interest in the case ofbreast or colorectal cancers. Two ap­ proaches were developed at the laboratory: the top-down SERological Proteome Analysis (SERPA) and the bottom-up MAPPing (Multiple Affinity Protein Profiling) strategies. The first one relied on two-dimensional electrophoresis (2-DE), immunoblotting, image analysis and mass spectrometry. The second approach deals with the use o f two-dimensional immuno-affinity chromatography, enzymatic digestion o f the antigens and analysis by tandem mass spectrometry.
