ABSTRACT

The NPM mutation cannot be detected by either standard cytogenetics or FISH and can only be identified using molecular techniques. Mutation of NPM can be inferred on IH C by localization of the protein in the cytoplasm of leukemic blasts (NPMc+), as opposed to the normal nuclear localization when un-mutated. Immunohistochemistry for cytoplasmic NPM may also aid in the evaluation for minimal residual disease. W hether or not IH C for NPM should be routinely done in all cases of AML-NK is still being debated.
