Development of a Robust, Environmentally Responsible Process for the Manufacture of Tofacitinib Citrate

Process development efforts that resulted in a robust, environmen-

tally responsible, commercializable process for tofacitinib citrate

will be presented in this chapter. The retrosynthetic strategy

to this potent immunosuppressant/immunomodulator, synthetic

approaches to key building blocks, and development of the end game

will be discussed.
