Protein array technology is readily amenable to different binding assays with various types of molecules, including DNA. It has become a powerful tool for genome-scale screening of protein-DNA interactions and for routine measurements of DNAbinding parameters of wild-type and mutant proteins. The advantages and main challenges of this emerging technology are described in this chapter. Ongoing developments in the field of the investigation of DNA-protein interactions and their relevance in biomedical research are discussed.
