ABSTRACT

Metabolome analysis is divided into two approaches: nontargeted analysis and targeted analysis.1 Nontargeted analysis covers all detected peaks by means of scan mode and utilizes both annotated and unannotated peak information for statistical analyses. This approach has been broadly applied to discovery-phase studies such as biomarker identification for medical

diagnostics. On the other hand, the targeted or widely targeted analysis hunts only “known metabolites” detected by single ion monitoring or selected reaction monitoring mode with the purpose of validating a biological hypothesis or machine-learning studies such as regression and discriminant analyses. In this chapter, we introduce the protocol for the nontargeted gas chromatography/mass spectrometry (GC/MS) based metabolomics.
