ABSTRACT

Ever since the original proposals that dopamine and its receptors could be involved in antipsychotic drug actions (1) and in the psychotogenic action of amphetamine (2), a debate about the possible role of this neurotransmitter has attracted a lot of attention. The dopamine hypothesis of schizophrenia was until recently supported only by indirect, pharmacological evidence. During the past decade this has changed, and there is now fairly convincing evidence of a dysfunction in the dopaminergic system in drug-naive schizophrenic patients.
