ABSTRACT
Samples of PPO extract were heated as described above and then were analyzed by polyacrylamide gel electrophoresis with 15 pi of each sample containing 7.5 |xg of protein applied to the gel. A mini vertical electrophoresis system (GIBCO-BRL, from Gaithersburg, MD) was used, leaving the process to run for 3 hours on six proteins of different molecular weight. They were then applied to another gel to obtain a regression curve of molecular weight versus Rf values. The proteins were developed with Coomasie blue R-250 and PPO activity of samples detected by incubating 30 minutes with 1% catechol.
