The hydrophobicity of an amino acid side chain is derived from the measurement of the solubility of the amino acid in water and an organic solvent.1,2 The data are converted into the molar free energy of transfer for the amino acid from an aqueous solution to a solution in the organic solvent at the same mole fraction at the limit of infinite dilution:
