Lysine, α-amino groups, tyrosine hydroxyl; preferred reaction is at lysine; pH 8 or greater; reaction can be "driven" a-amino groups at pH less than 6.5. Avoid nucleophilic buffers such as Tris; hydrolysis of the reagent is an issue above pH 9.5. Acetic anhydride has been used for trace labeling in the study of protein conformation and more recently the deuterated derivative has been used in proteomics for differential isotope tagging
