The intent of this chapter is to review the use of a culture-based strategy to prepare autografts for patients with chronic myeloid leukemia (CML). As with other in vitro techniques, the goal is to minimize the content of leukemic stem cells in the autograft while retaining sufficient Philadelphia chromosome (Ph)-negative stem cells to enable prompt restoration of blood cell formation.
