ABSTRACT: It was considered that 1-acetyl-6,7-dihydroxyl-1,2,3,4-Tetrahydroisoquinoline (ADTIQ), and its precursor methylglyoxal were closely related to glucose metabolism. ADTIQ was similar in structure with catecholamine isoquinolines (CAIQs). Therefore, ADTIQ was thought to be related to diabetes and Parkinson’s Disease (PD). The quantitation of ADTIQ in a biological sample could play an important role in detection of diabetic-PD. Herein, a sensitive and precise method was established to quantify ADTIQ by LC/MS/MS in Multiple Reaction Monitoring (MRM) mode. The [M + H]+precursor/ product ions were 208.1/190.1. The linear range of calibration curve was from 0.01-2 nM, and the lower limit of ADTIQ quantitation is 0.002 nM. Recoveries for biological sample were 137-162% with acceptable inter-day, intra-day precision and accuracy. Furthermore, the assay method was used for the determination of ADTIQ in rat substantia nigra.
