ABSTRACT

Naturally, advanced material civilization resulted in the destruction of biological resources, causing a variety of emerging viruses since late 20 Century. Principle strategy against numerous communicable viral diseases should be prepared suitable vaccine under the following four conditions: first, highly effective; second, urgent short period; third, large scale production; fourth, low cost. Silkworm-derived Virus-Like Particle (VLP) vaccines may accept the above four conditions. Even though we have compared the yield of vaccine protein (hemagglutinin: HA), it appeared that the level of hemagglutinin production in silkworm was influenced by characteristic viral genome and insertion of codons specific silkworm. For example, although when used non-codon optimized wild type H5 HA gene in the expression system, its titer was approximately 10,000, the use of codon-optimized target gene increased its HA titer to 50 - 100 times higher. The above evidence was obtained from H5 Fukushima recombinant. Similarly, recombinant Korea H7 – BmNVP could produce 400,000 HA titer with use of codon optimized H7 Korea HA gene. The codon optimization effect was also confirmed by the level of m-RNA synthesis.
