ABSTRACT

A multitude of selection and screening strategies for phagedisplayed libraries have been developed since the technique was first described by Scott and Smith (1). Each relies on the ability to discriminate desired phage from the library either through selective capture to allow the removal of unbound phage or through selective elution, as in the case of substrate phage to specifically elute desired phage (see Chapter 7). Ultimately, the degree to which target directed phage can be separated from the rest of the phage library will determine the effectiveness of the panningmethod. This chapter summarizes many of these strategies and discusses the considerations that must be kept in mind when using them.
