Purification of sufficient numbers and amounts of proteins is commonly thought to be one of the more important steps in high-throughput structure determination. In recent years, many groups have honed methods to produce purified prokaryotic proteins in large scale. As international projects inevitably move to focus on eukaryotic and membrane proteins, there are new requirements to develop cost-effective methods to produce these proteins. This review will describe existing methods for producing bacterial proteins, some new strategies to produce bacterial proteins more economically, and approaches that might be used for eukaryotic and membrane proteins.
