ABSTRACT

Understanding the pharmacokinetics and the differences which occur in children and adolescents helps to explain the possible problem areas and how potential drug-drug interactions can take place.

Absorption Orally administered drugs can undergo a number of interactions during their passage through the gastrointestinal tract. The important factors that affect absorption are similar in adults and children over 1 year. They include: the gastrointestinal pH, which alters the lipid solubility of drugs and their subsequent diffusion across mucus membranes; the formation of large drug complexes which are then poorly absorbed; and factors that alter gut motility and as a result either enhance or delay gastric emptying.
