APP. Since the APP 5′-UTR encodes both IL-1 and iron regulatory elements, we proposed that APP is intimately involved in iron metabolism in health.5 We also developed an RNA-based screening protocol for the selection of drugs that interact with the APP 5′-UTR to control APP holoprotein levels and hence Aβ output.2,7 Here we discuss how the acetylcholinesterase inhibitor (AChEI) phenserine, and the M1 muscarinic agonist AF102B, influence APP gene expression through iron-and IL-1-specific pathways.
