ABSTRACT

To determine the effect of enzyme concentration on the catalytic activity, a graded amount of enzyme is added to the reaction mixture and the lipase activity is assayed by the titrametric method. The optimum temperature for the catalytic activity of lipase was assayed at the temperature range of 10-50 °C using tributyrin as the substrate. The catalytic activity of KBS-plip1 lipase was determined in the presence of chelating agent Ethylenediaminetetraacetic acid (EDTA) and emulsifying agent gum arabic. Thermal stability and activity of the KBS-plip1 lipase enzyme were estimated at different temperatures. The stability of the organic solvent of purified lipase is studied by preincubating the purified enzyme with the various organic solvents viz methanol, ethanol, 1-propanol, 2-propanol, glycerol, acetone, acetonitrile, DMSO, and benzene at 37 °C for 120 min. To check stability and activity of KBS-plip1 lipase in the presence of organic solvents, the assay was performed with the different organic solvents.
