I TRICYCLIC ANTIDEPRESSANTS (TCAs) A IN GENERAL (a) Mode of action Monoamine reuptake inhibitors (MARIs) - inhibit the reuptake of both serotonin and noradrenaline into the presynaptic neurone, with the result that both neurotransmitters accumulate within the synapse. Such biochemical changes occur within several hours following administration of the drug, while the antidepressant action of the drug is delayed for about 2 weeks, indicating that some secondary process must be taking place.
