ABSTRACT

This chapter discusses clorazepate, a neuropsychotropic drug. It presents the approved indications of the drug for neuropsychological disorders, usual dosage and administration, available dosage forms, storage, and compatibility, and the mechanism of action. Information on relative contraindications, clinically significant drug interactions, adverse drug reactions, and overdosage is also provided. Sold under the trade names Gen-XENE®, Novo-Clopate®, Tranxene®, and Tranxene-SD®, clorazepate is prescribed for partial seizures and those associated with the acute alcohol withdrawal syndrome. The exact mechanism of action of clorazepate has not yet been fully determined. However, it appears to be mediated by or to work in concert with the inhibitory neurotransmitter, GABA. Concurrent alcohol use may significantly increase the CNS depressant action of clorazepate. Patients should avoid or limit their use of alcohol while receiving clorazepate pharmacotherapy.
