Dikaryoticisolatesofthewood-deterioratingbasidiomycetesaremaintainedon2%maltextract agarmedium(MEA)orpotato-dextroseagar(PDA)inthedarkat27°Cand70%relativehumidity.
Thefollowingmediacanbeusedforfruitingbodyformationandbasidiosporeproduction: completeplusyeastextractmedium(CYM), 11sawdustmedium,5PDAmedium(Difco),MEA medium,andachemicallydefinedmedium2containingglucoseorWalsethcellulose12andammoniumtartrate.Anadditional25mMKH2P041isaddedtothebasalmedium,BIII6tothechemically definedmedium.Pyrex®storagedishes(ComingNo.3250)containing50mLofthecomplex mediumorchemicallydefinedmediumareinoculatedatthecenteragarofplateswitha6-mm mycelialdiscobtainedfromafreshculture.Plasticpetridishescontaining20mLofthesame mediumcanbeusedinplaceofthestoragedishes.
