In addition to separating testosterone from its esters, chromatographic separation removes both components of the oil which interfere with the spectrophotometric measurement. The separation of analyte can be done most simply and effectively by means of one of the chromatographic techniques. Column chromatographic separation of organic compounds is used primarily in preparative chemistry. Column chromatographic separation for analytical purposes, although its significance has been reduced with the spread of thin layer chromatography , is still an important method of pharmaceutical analysis, for the measurement of both drug preparations and biological samples. Consequently, in a very short time a very efficient separation can be performed, and by the continuous detection of the eluate a chromatogram, also excellent for quantitative analysis is obtained. Another very important application field of liquid chromatography–diode array UV detector combination is the investigation of the homogeneity of chromatographic peaks.
