ABSTRACT

The bacterial cell wall provides a rigid structure and shape to the bacterial cell. The bacterial cell wall also helps to differentiate bacteria into Gram-positive or Gram-negative groups based on the cell wall composition. The cell wall also possesses the antigenic properties. The cell wall-related compounds produced by phytopathogenic bacteria play an important role in the induction of disease water-soaking reaction in the host plant. The bacterial cell wall is critical for the determination of cell shape during growth and division, and maintains the mechanical integrity of cells in the face of turgor pressures several atmospheres in magnitude. A straightforward procedure for isolation and preparation of bacterial cell wall for biological analysis of peptidoglycan is ultra performance liquid chromatography. The bacterial suspension is centrifuged at 19000 rpm for overnight in ultracentrifuge at 4°C and the supernatant is filtered through a Millipore filter. The filtrate is precipitated with ammonium sulfat.
