ABSTRACT

Non-selective inhibition of both isoenzymes is a feature of the NSAIDs and relatively specific inhibition of the type 2 (inducible) enzyme is a feature of the COXIBs.

Uses and efficacy: acute pain Many NSAIDs have been trialled in a standard acute pain model (e.g. following dental extraction), and found to be effective. The number needed to treat (NNT) for single doses is well established in controlled trials: diclofenac 50 mg, NNT = 2.3; ibuprofen 400 mg, NNT = 2.4.
