ABSTRACT

The chemical approach may involve the design and development of novel drugs synthetically. The prodrugs of haloperidol were synthesized and evaluated for their hydrolysis in simulated environment corresponding to liver and small intestine. The prodrug of acetaminophen with proline was designed to mask the bitter taste of acetaminophen, which is due to the phenolic group. A number of drugs have amino groups in them. The amino group may be present in the form of primary, secondary or tertiary amine or heterocyclic amine. The amino groups containing drugs got ionized at physiological conditions. The peptide prodrug of glucosamine with more gut permeability through gut peptide transporter-1was synthesized. The concept of prodrug has resulted in giving a lot of drugs in market which has clinical applications. The rationale of developing a new drug may involve the study of specific receptor or enzyme that involve in pathological condition.
