ABSTRACT

Among various strategies to increase monoclonal antibody (MAb) production, suppressing cell growth so that hybridomas can divert their energies to MAb production is perhaps one of the more simple and novel strategies. We have shown that eRDF medium (enhanced RDF, which consists of a combination of RPMI:DMEM:Ham's F12 in the ratio 2:1:1, commercially available from Kyokuto Pharmaceutical Industries Co., Japan) is a superior medium for MAb production [1], and that by increasing the osmolarity of a medium and adding sodium butyrate, hybridomas can be stimulated to overproduce MAb [2], This response has been demonstrated in various cell lines [3], Cells under such stresses also increase their specific metabolic activity, total RNA content, and 102the transport of amino acids, whereas growth and total DNA content are concomitantly reduced [4].
