ABSTRACT
Immunohistochemical detection of proliferating cells in regenerating Cuvierian tubules of Holothuria forskali was performed after intracoelomic injection of 200 mg bromodeoxyuridine (BrdU), a thymidine analogue. After digestion of the sections with trypsin and acid hydrolysis, BrdU-labeled nuclei were revealed by the immunogold-silver staining (IGSS) procedure. Among the several fixative tested, Bouin-Allen mixture appeared the most performant and gave clear-cut labelings of S-phase cells in regenerating tissues.
