ABSTRACT

In any project involving quantiŽcation of ªuorescence, the Žrst consideration is which sort of microscope to use. The confocal microscope is probably the most accurate tool available for measuring intensity within a deŽned voxel space, but we need to be clear about what this implies. If we want to measure the total ªuorescence of an organelle-for example, to determine the DNA content of a nucleus-voxelbased measurement is not really what we want, and wideŽeld ªuorescence or ªow cytometry is more likely to be a useful tool.
