Before use, each lot of C ' should be tested for specific (antibody mediated) and nonspecific (direct) cytotoxicity. Rapid, up-front screening on leukocytes may be performed using assays that are in place in most tissue-typing labora­ tories. Sources that pass these tests should then be tested in the specific system in which they are to be used. Target destruction and normal cell recovery and functional activity should be assessed using the tests that will ultimately be used to optimize purging efficacy. For nucleated target-cell lysis applications, the most widely used tests are limiting dilution analysis (8,38) and 51Cr-release cytotoxicity assays (43).
