ABSTRACT

A first screening of culture supernatants should be done as soon as clones appear and cell damage should be avoided well before the medium turns to bright yellow. However, controls must be carried out in order to detect polyclonal-specific antibodies secreted by unfused splenocytes. A second screening after two or three changes of culture media is often necessary. The chosen method must allow a rapid selection of all interesting hybrids, and hence has to be simple and sensitive. Methods used in our laboratory for the detection in the culture supernatants of monoclonal antibodies (MAbs) against soluble antigens are discussed later in the chapter.
