I. INTRODUCTION A. Regulation of Glucose Homeostasis The maintenance of whole-body glucose homeostasis involves the interplay between the disposal of blood glucose in the periphery, primarily in skeletal muscle, and the delivery of glucose to the blood, either endogenously from the liver (hepatic glucose production) or exogenously from dietary sources (Fig. 1A). Disruptions in the normal regulation of either process-hepatic glucose production or skeletal muscle glucose disposal-will lead to chronic elevations of blood glucose and the development of diabetes mellitus (see Fig. 1B).
