ABSTRACT

Figure 1 Permeability (Papp) of model chemicals in Caco-2 cells cultured as the conventional 21-day culture and as 3day culture using a proprietary protocol developed in our laboratory. The chemicals chosen are known to permeate through Caco-2 monolayer cultures transcellularly (testosterone), via active transport (glucose), and paracellularly (propranolol and mannitol). Vinblastine is a Pgp substrate and verapamil is a Pgp inhibitor. The enhanced permeability of vinblastine in the presence of verapamil therefore reflects the presence of Pgp in both the 21-day and 3day cultures. The transepithelial electrical resistance (TEER) values of the two cultures are also presented.
