ABSTRACT

The lipidic prodrug approach that resulted in lymphatic transport includes a number of drugs; some examples are testosterone, L-dopa, and bexaroten. Special liker design and drug delivery system can significantly improve drug delivery; lipid-based conjugates and drug delivery systems can form hydrophobic interactions, stabilizing the drug loading and overall stability of the drug formulation. Resistance to anticancer agents is a rising problem, hence an innovative approach needs to be used, for instance drugs in an enzyme-instructed self-assembly. phospholipids-based prodrug approach was shown to be very useful tool for cancer therapy, and it represents a rather novel tool for drug targeting to the cancer cells. Lipidic prodrugs can influence the safety profile of the parent drug and minimize adverse effects. A combination of a lipid-prodrug approach and different formulation strategies can largely enhance drug delivery and targeting. The modern prodrug approach aims to target specific transporters or enzymes, providing site-specific drug release.
