ABSTRACT

In this chapter, we present different methods for making libraries of recombinant polypeptides. First, we describe how to create highly defined libraries by using synthetic DNA. Second, we present alternative approaches that can be used to introduce mutations randomly. In these cases, the position and nature of the mutations are not defined by the experimenter. Both types of methods enable the production of libraries that can be used to select polypeptides with desired functions. Finally, we describe techniques that have been developed to

further increase the diversity of these libraries by either recombination or shuffling.
