ABSTRACT

N-glycosylation is one of the most common and important post-translational modifications of proteins. Practical glycome qualitative and quantitative methods are crucial for the fields of disease marker discovery, protein glycosylation function research and quality control of biopharmaceuticals. Mass spectrometry has become one of the most effective tools for N-glycome analysis due to its high sensitivity and resolution. This chapter will describe major methods and improvements in sample pretreatment protocols and instrumental analyses for N-glycome analysis, mainly including N-glycan release, purification and enrichment, separation, derivatization and some common qualitative/quantitative analysis strategies.
