ABSTRACT

This chapter covers the downstream processing (DSP) of viral‐based vaccines. The chapter provides an overview of the traditional purification methods that have been used, until the current trends. The content includes all the unit operations used in bioprocessing (clarification, intermediate purification, and polishing), explaining the fundamentals of each step. It discusses the overall goals from the standpoint of purity, recovery yield, and efficiency as well as the strategies employed to achieve those goals. The entire section is completed with the recent advances in commercially available materials as well as examples of typical viral‐based purification processes with its respective outcomes. Furthermore, this chapter also highlights the importance of robust analytical tools for in‐process control and process development. Finally, the reader will also have an overview of the future of purification technology through process intensification.
