ABSTRACT

This chapter presents the local BOIN design of Liu and Yuan. It discusses the escalation and deescalation boundaries used by the BOIN design. The Bayesian Optimal Interval (BOIN) Design is a statistical methodology for designing a dose finding clinical trial where the goal is to identify the maximum tolerated dose of the investigational medicinal product. The BOIN design was proposed by Liu and Yuan. The general form of the BOIN design can be described via the decision rules used by the design once the trial is in progress. The trial is conducted such that patients are treated in cohorts with the sample size of each cohort prespecified.
