ABSTRACT

Gentiooligosaccharides are a new kind of oligosaccharide, which is a promising product of high value-added oligosaccharides. Compared with plant extraction and chemical synthesis, enzymatic preparation of gentian oligosaccharides have the advantages of mild reaction conditions, low energy consumption, low cost, easy separation, and purification. At present, gentian oligosaccharides can be realized mainly by transglycosylation or reverse hydrolysis synthesis catalyzed by glycosidases with β-1 amino acid 6-glycosidic bond synthesis activity. In this chapter, we introduce the function and application of gentian oligosaccharides, and summarize the research progress in the preparation of β-glucosidase and β-1mai 6-glucanase by gentian oligosaccharidase, including the source, properties, structural mechanism analysis, and engineering modification of these enzymes. In addition, this chapter also reviews the single-enzyme or two-enzyme synergistic reaction system and its optimization for the synthesis of gentian oligosaccharides catalyzed by these two kinds of enzymes.
