ABSTRACT

Solatol is a noncardioselective beta-adrenoceptor blocking agent with Class III antiarrhythmic properties. The most important symptoms after overdosage (2.4 g to 8 g) are hypotension, bradycardia, and a prolonged Q-T interval, with the possibility of torsades de pointes. In this chapter, the authors develop a rapid HPLC method for the determination of concentrations of sotalol in serum. They measure serum sotalol concentrations and Q-T intervals in a patient after acute sotalol poisoning. Serum sotalol concentrations were determined by HPLC on a Chrompack 10 cm × 3 mm column filled with Hypersil ODS 5 um. Since sotalol is not protein-bound, the authors use ultrafiltration for sample preparation. The absolute recovery is higher than 90%, and the method has a good precision.
