FluorescencePolarization.Fluorescentsolutionswhichareexcitedwithverticallypolarizedornaturallightemitpartiallypolarizedfluorescenceasviewedatrightanglestotheincident beam.Polarizationoffluorescenceisduetothefixedrelationshipbetweenmolecularorientation andabsorptionandemissionoffluorescence.IfasmallfluorescenthaptenisboundbyanAb, therewillbemarkedreductionintherotationalfreedomofthehaptenandacorresponding increaseinpolarization.Again,itispossibletoobtainrelativevaluesoftheextentof fluorescencepolarizationfor100%unboundAgandtocomparethesewiththeextentof polarizationforboundhaptensandtherebyestablishequilibriumconstants(28,40,41).The commonlabelingreagentsforproteinAgsarefluoresceinisothiocyanate,rhodamine,and dimethylaminonaphthalenesulfonylchloride.
