ABSTRACT: In order to overcome some shortage during traditional fermentation such as decrease of cell activity, low volumetric productivity and product inhibition, alginate-chitosan (AC) microcapsules encapsulated genetically engineered strain GS115-pPIC9k-ChiA4.0 were developed. Microcapsules with different chitosan molecular weight and different membrane formation time were prepared. Results showed that cells grew well in the AC microcapsules. The cultured GS115-pPIC9k-ChiA4.0 inAC microcapsules which modified with 100 kDa molecular weight chitosan and 10 min formation time showed the highest OD600 value (9.255) and the enzyme activities (37.5 U) respectively. Interestingly, when the molecular weight of chitosan decreased or the formation time increased, both the cell growth rate and enzyme activities of GS115-pPIC9k-ChiA4.0 in AC microcapsules decreased. Thus, the AC microcapsules we prepared were suitable for immobilizing GS115-pPIC9k-ChiA4.0 cells and will show a promising application in the field of cell immobilization.
