ABSTRACT

ABSTRACT: In this study, the proliferation conditions of recombinant Escherichia coli BL21 (DE3) / pET-28b-aroGM175 which produced L-tryptophan were optimized. The effects of temperature, loading volume and pH on residual glucose and density of recombinant strain were optimized by response surface methodology (RSM). Finally, the study found that the optimal proliferation condition obtained by RSM was as follows: temperature of recombinant strain Escherichia coli BL21 (DE3) / pET-28b-aroGM175 was 34.75◦C, loading volume was 150 mL medium of 1000 mL bottles and initial pH of culture was 6.955. Thus this study provided a scientific basis for large-scale fermentation production of L-tryptophan by using genetic engineering strain.
